Missions
The Mitolab team specializes in exploring mitochondria and their functions in physiological and pathological contexts. Several projects focus on mitochondrial DNA and the pathological consequences of its instability (mutations or depletion). As part of a project funded by the French National Research Agency, we are seeking to characterize the therapeutic potential of mitochondrial-targeted nucleotides to treat mitochondrial DNA depletion syndromes. The recruited engineer will determine the impact of these molecules on mitochondrial functions and health in in vitro and in vivo models mimicking these depletion syndromes.
Activities
– Cell culture experiments (fibroblasts and commercial cells) with treatments
– Analysis of mitochondrial energy efficiency (oxygraphy, ATP dosage)
– Cell imaging: labeling with probes and use of microscopes (confocal and transmission electron microscopy)
– Molecular biology experiments: quantitative PCR, Western blot, BN-PAGE
– Manipulation of rodents and administration of pharmacological treatment
– Maintain an up-to-date laboratory notebook, write protocols
– Processing and analysis of results
– Presenting results in the form of experimental reports and oral presentations
– Ensure scientific and technological monitoring
– Provide research support (stock and database management, orders, receptions)
SKILLS
– Excellent skills in cell culture
– Mastery of basic techniques in molecular biology
– Diploma in animal experimentation (level 1 or 2)
– Ability to work in a team and independently
– Compliance with hygiene and safety rules

Work context
The Mitolab team is one of two teams that make up the Mitovasc research unit based in Angers, within the university hospital center. It is a team of around fifty people, researchers, ITA clinicians, and students, with several research areas dedicated to mitochondria. The engineer will join the “mitochondrial genome instability and aging” group and will benefit from a privileged environment for carrying out their tasks, with access to cellular and molecular biology, microscopy, and animal facilities, managed within the federated research structure ICAT 4208.
Constraints and risks
Risks associated with laboratory work (biological agents, chemicals, handling rodents). The engineer will undergo training in risk prevention.
