    I am highly motivated to work in the field of T cell activation and differentiation. During the course of my graduate work, I have developed an interest in the inflammation and pathways regulating T cell function. I am especially intrigued by T cell signaling and how different signals TCR, cytokine signaling and co-receptors like xxx and TLR work together to induce T cell proliferation and differentiation. How Notch regulates xxx signal strength by regulating E3-Ubiquitin ligases and thereby modulates T cell differentiation. Kinetics of NF-κB signaling in T cells and role of Notch signalling in the formation of lipid raft and immune synapse are other aspects which excites me.

    My graduate work has focused on the immune dysfunction in oral cancer and role of immunosuppressive factors/cells present in the tumor microenvironment. I have worked on the lymphoid as well as myeloid cells during the course of PhD. I have analyzed the different regulators (positive as well as negative) of T cell signaling and how they modulate the T cell function in cancer setting. The important among them was the Notch signaling which has implications in increasing the efficacy of T cell based therapy. We have addressed the root cause of immune dysfunction and its association with MDSCs, Tregs and Th17 cells. We have addressed how MDSCs accumulate, their life span, suppressive potential, and also how they induce Th17 differentiation and inhibit Treg differentiation in in vitro conditions.

    Apart from this I have also worked on project “xxxx signaling in regulating effector functions of xxxx cells. I am well versed with the biology and in vitro expansion of xxxxx cells.

    During my doctoral program I have thoroughly used multicolor flow cytometry (nine color), western blotting, real-time PCR, siRNA, molecular cloning, co-culture assays, Mixed leukocyte reactions, Immunomagnetic sorting of cells (MDSCs, Tregs, Naïve CD4 cells and CD3 cells, γδ T cells), cytometric bead array and ELISA for quantitation of cytokines, proliferation assay (Dye dilution and tritiated thymidine assay), 51Cr release assay, confocal microscopy, chemotaxis assays to perform the experiments.

    I have learned animal handling especially animal dissection, harvesting of liver, spleen, mammary glands, vertebrae and femur bones, Tail vein injection.

    Summary of Project#1:

    Phenotypic heterogeneity allows individual cells to behave different from the bulk. This non-genetic variability provides a critical route to therapeutic resistance and relapse of cancer. We identified rare, slow cycling, dynamic, pre-existing drug tolerant state in xxx-Cell xxxx. Single-cell analysis revealed a strong inherent drive to re-program daughter cells to occupy the xxxx state. Transcriptional reprogramming in resistant phenotype, in particular xxxxx helps in maintenance of an equilibrium between the drug-resistant and drug-sensitive cells.     Summary of Project#2:

    Metabolic flexibility allows cells to adapt to various environments and limits efficacy of metabolic drugs. Therapeutic targeting of cancer metabolism relies on defining limiting requirements and vulnerabilities in the highly dynamic metabolic network of cancer cells. We characterized metabolic reprogramming in response to pharmacological inhibition of mitochondrial complex xxx in malignant xxxx lines, xenograft models, and isolated xxx cells. Metabolic adaptation to complex xxxx dysfunction is mediated by mitochondrial reactive oxygen species (mROS) serving as a stress signal that activates xxxx. Inhibition of the xxxx/xxxx axis selectively disrupts metabolic adaptation and survival during complex xxx dysfunction in malignant xxxx, xenograft models, acute lymphoblastic leukemia (ALL) and chronic lymphocytic leukemia (CLL) patient’s samples (manuscript under communication).

    Emerging evidence indicates that cellular metabolism is a critical factor in cancer progression. We find that malignant xxx and xxx cells oscillate between non-invasive and highly invasive states providing a rationale for novel treatment approaches “locking” cells into a non-invasive state. The highly invasive state shows a reprogrammed metabolic profile with xxx xxx and xxx xxxx indicating a critical role of bioenergetic wiring in disease dissemination.     Research Experience:

    Summary: Oral squamous cell carcinoma is often associated with a lymphocytic infiltrate that is believed to mount an in vivo immune reaction to the tumor cells. The extent of the lymphocytic infiltrate has been correlated with clinical prognosis, suggesting the relevance of the immune response to tumors. Accumulating evidence indicates that a dynamic cross-talk exists between tumours and the immune system that regulates tumour growth and metastasis. The focus of the present study is to identify factors secreted by oral tumor that affect key signaling events in lymphocytes and also to understand the role of myeloid-derived suppressor cells (MDSC) in impairing immunity in oral cancer patients.

    xxxxxxxxx:

    Part I: Extracellular xxxx mediates TCR xxxx downregulation via xxx activation in oral cancer.

    Observations:

    • Oral tumor-derived factors promote immune suppression by downregulating xxxxx expression without affecting other key T cell signaling molecules
    • xxxxxxxxx (xxxx) was identified by the proteomic approach and our results established a causative link between xxxxx downregulation and xxxxx stimulation.
    • This effect was associated with only xxxx and not with other member of family xxxxx and oral tumours were expressing xxxxxat both protein and mRNA level.
    • Incubation of xxxx cells with cell-free supernatants of oral tumors or recombinant human xxxx (xxxx) induced xxxx activation which resulted in xxxx downregulation.
    • xxxx inhibition/downregulation using pharmacological inhibitor or siRNA restored downregulated xxxx expression in xxx cells induced by cell-free tumor supernatant or xxxx.
    • Collectively these results show that xxxx leads to impairment in xxxx expression and might be inducing xx cell anergy by interacting with xxxx, thus offering an explanation that might be applicable to xxxx deficiency observed in cancer and diverse disease conditions.

    Part II: Defining the role of xxx in down regulation of xxxx cell signaling molecule xxxxx in oral cancer patients

    Observations:

    • xxxx expression is upregulated compared to the xxx in xxx cells of oral cancer patients compared to healthy individuals and serum levels of xxxx are higher in oral cancer
    • Induced xxxx expression on xxx cells made them susceptible to the xxxxstimulation.
    • xxx resulted in the selective decrease in the xxxx expression in xxxx cells and blocking of xxxx abrogated xxxx induced downregulation of xxxx.
    • Elevated reactive oxygen species levels and xxxx was observed upon xxx stimulation.
    • xxxx altered xxx signaling as observed by altered xxflux and decreased xxx

    Part III: “Inhibition of xxxx signaling alters xxxx expression in human xxxx cells by regulating xxxx ligases.”

    Observations:

    • Effector functions of xxxx cells driven by xxx cell receptor and antigen non-specific signals  are known to be regulated by xxxx signaling
    • Human xxx cells have differential expression of xxx receptors, ligands, and target genes which get activated on stimulation with xxxx.
    • Inhibition of xxxx signaling appeared to regulate activation of xxxx cells and affect proximal xxxx cell signaling by regulating xxxx expression.
    • Inhibition of xxxx signaling affected protein levels of xxx by inducing the expression of an xxxx ligase.
    • Inhibition of xxxx signaling induced xxxx cell anergy as depicted by increased xxx, xxxx expression and altered xxxxx flux.
    • Absence of xxxx signaling, xxxx induced activation of xxxx cells and xxxx production was down-modulated which was evident at decreased xxxx and xxxx expression.
    • The absence of xxx signaling prevented proliferative responses despite strong signaling through xxx and xxx receptor in a xxx dependent manner. This study shows how different signals cooperate with each other to support proliferation and activation of human xxxx

    xxxxxxxxx.

    Part IV:  Contribution of xxxx derived suppressor cells, Regulatory xxxx cells and xxxx cells in the inflammatory tumour microenvironment of oral cancer patients.

    Observations:

    • Monocytic xxxs were prevalent in periphery while granulocytic subset was dominant in the tumor compartment.
    • Mo-xxxxs were lacking in the tumour compartment and were replaced by tumour associated macrophages under the influence of oral microbiota, hypoxia and heat shock proteins (HSPs). MDSCs responded to these factors via toll like receptors (TLR2 and TLR4). Tumour microenvironment was enriched by xxxx and xxx positive macrophages.
    • MDSCs accumulation was not associated with resistant to apoptosis as they were undergoing higher turnover compared to their mature counterparts. They expressed higher levels of xxx and xxxx. Apart from this MDSC expressed higher levels of DR5, cleaved caspase 3 and p66shc. Accordingly the levels of ROS secreted by these was drastically high which might be regulating the emergency myelopoiesis and thereby accumulation of MDSCs in peripheral blood.
    • MDSCs suppressed T lymphocyte proliferation, decreased xxxx xxxx expression and IFN-γ production and expressed higher levels of COX-2, arginase 1, xxxx and Hes1. Effect was more pronounced on xxxx cells. xxxx cells were producing decreased levels perforin and granzyme B and were of exhausted phenotype.
    • IL-6 induced xxxx in MDSCs regulated PDL1, xxxx and xxxx expression. Oral cancer Mo-MDSCs expressed the xxx and xxxx which might be the outcome of higher pSTAT3 in these cells.
    • xxx which might facilitate crosstalk between MDSCs and regulatory T cells (Tregs) and increased Tregs were observed in OC patients and correlated with stage of the tumor and xxx levels produced by MDSCs.
    • Tregs were accumulating in the tumour compartment and we found that compared to xx and xxxx, Tregs from tumour compartment patients expressed higher levels of xx and xx. Oral tumour derived cells also expressed xxx and xx chemokines. Chemotaxis assay confirmed that Tregs have higher tendency to migrate to the tumour site via xxxxx axis thus creating an immunosuppressive environment.
    • In-vitro data showed that MDSC inhibit the TGF-β driven generation of iTregs from naïve xxx T cells suggesting that Treg accumulation might been occurring independent of MDSCs.
    • Cytokine profile of MDSCs had ability to alter differentiation of xxx naïve cells to Th17 cells and MDSCs significantly enhanced the in vitro generation of xxx cells. Consequently TH17 cells were higher in oral cancer patients suggesting that increased frequency of MDSCs and Tregs results in T cell tolerance and induce a chronic inflammatory state facilitating tumor growth. These cells thus could be used as a potential targets to modulate antitumor immunity.

    Title:  “Role of xxx in regulation of xxx lymphocytes functions.”

    Summary: xxx cells recognize phosphorylated antigens and play an important role during infections and in tumor immunity. The aim of the present study was to investigate the role of Notch signaling pathway in antigen specific responses of xxx cells. xxx cells were found to be predominately expressing xxx and xxx genes. The presence of active Notch intracellular domain in γδ T cells was confirmed by western blotting. Proliferation of xxx cells and IFN-γ release in response to xxx and xxxx stimulation was inhibited in the presence of GSI. Diminished lysis of tumor targets by γδ T cells as shown by 51Chromium release assay in the presence of GSI or by specifically knocking down the Notch genes, confirmed that Notch is involved in cytotoxic effector functions of xxxx cells.

    xxx-xxxx (MSc dissertation) xxxx University (Supervisor: Dr. xxxx)

    Summary: Pxxx is an important component of apoptotic response and life span determinant. xxxx, a xxxkda protein, has been identified as a splice variant of xxxx, but unlike these variants, which are involved mainly in the transmission of mitogenic signals, xxxx functions in the intracellular pathway by converting oxidative signals into apoptosis. However, recently in human carcinomas, xxx has been found to be involved in regulating their proliferation. Elevated expression of p66shc protein is observed in prostrate carcinomas, esophageal carcinoma as well as in all kinds of proliferating thyroid tissues. But its role in human carcinogenesis has not been thoroughly dealt with. Hence, its expression in cancers needs to be thoroughly investigated which may help in understanding the signal transduction pathway operating in cancers. We extended our work on role of p66shc expression in colorectal and bronchogenic carcinomas. Our data support for a novel cancer-specific role of p66shc in colorectal and lung carcinomas

    Summary of Doctoral work:

    Fanconi anemia (FA) is a genomic instability and cancer predisposition syndrome. FA proteins mainly xxx and xxx have pivotal role in DNA intercrosslink repair. However, structure, functions and interactions of these proteins are not well understood. Therefore, in my Ph.D Thesis, I demonstrated the structural and functional role of human xxxx repeat (cite here with published work or under communication/preparation). I investigated the structural and functional properties of xxxx domain and its biologically relevant mutants (xxxxet al. manuscript under preparation). Furthermore, I performed the interaction studies of the functional domains present in the xxx, xxxx and xxxx proteins (cite here with published work or under communication/preparation).     1. To find out the proteins interacting with xxx repeats sequence of xxx gene using CRISPR-based method.

     

    Introduction: xxx gene is present in chromosome xx of human which codes for mitochondrial protein that regulates mitochondrial iron transport and respiration. Increase in number of xxx repeats in its intronic region leads to development of xxxx’s xxx disease which is a neurodegenerative disease. Increase in number of xxx repeats lead to decrease in xxx expression. In unaffected person, xxx expression is maximum when one of the xxx copies is affected. FXN mRNA levels decrease at some extent and when both the copies are affected, xxxx mRNA levels completely decreased. Briefly, describe about the previous work done in our lab. We used synthetic transcription elongation factor to selectively activate xxxx expression in xxxx patients. xxxx is basically composed of two elements: one is polyamide that binds to xxx repeats of xxxx gene and other is xxx which recruit elongation machinery to the site where it is bound. The crystal structure of polyamide nucleosome complex and xxx-xxxx complex designed to identify appropriate site for conjugation. After conjugating, we inserted this xxxx in xxx patient cells. The polyamide fragment in this xxx will bind to xxx repeats and attached xxx. xxx will then bind to xxx and then will recruit elongation machinery to the site and allow transcription to occur. xxxx mRNA is then transcribed. This leads to rise in xxx mRNA expression.

    1. Insertion of Nano luciferase (Nluc) and Enhanced Green Fluorescent Protein (eGFP) gene into the xxx exon of human xxxx gene using CRISPR/ Cas9 genome editing method.

     

    Intoduction: xxxx xxx is the most common inherited disease that causes progressive damage to the nervous system. This disease is caused by the genetic mutation (expansion of an intronic xxx triplet repeat in the xxx gene) leads to reduced expression of the mitochondrial protein xxx. It can also lead to scoliosis, heart disease and diabetes, but does not affect cognitive function. The disease is progressive, and ultimately a wheelchair is required for mobility. Its incidence in the general population is roughly 1 in 50,000. Here we demonstrate the creation of robust xxxx reporter or xxx reporter lymphobalstoids/Fibroblasts cell line by knocking in a Nano luciferase (Nluc) or Enhanced Green Fluroscent Protein(eGFP) gene into the human xxx gene (using CRISPR/Cas9 genome editing method. These reporter line is a good resource for drug screening as well as testing potential genetic reactivation strategies.
