The goal with the project is to characterize important molecular mechanisms in the airway epithelium during inflammatory reactions. The project assignment includes to develop the use of a cell culture system where stem cells isolated from murine tracheas are cultured at air-liquid-interface to recapitulate a native lung epithelium. The cell cultures will be stimulated with different inflammatory signals and extracellular vesicles and mucins will subsequently be isolated for further analysis. Extracellular vesicles and mucins will be analyzed using methods such as flow cytometry, mass spectrometry, western blot and RNA analysis by qPCR and sequencing. Experimental mouse models of asthma will be used for analysis of extracellular vesicles and mucins produced in vivo in the lung tissue. In these experiments, detailed studies of inflammatory cells will be done by flow cytometry and inflammatory signals will be analyzed by ELISA.

The research project is aimed at the development of improved biomarkers for tau, amyloid and other neurodegenerative pathologies in the brain.
