Perceptual discriminations are among the most important decision making processes. One of the most prominent questions in systems neuroscience is how the perception of a given stimulus is encoded in the brain and how this translates to a learned decision that is followed by a motor response. Using the repertoire of experimental techniques available in our lab, such as two-photon and three-photon imaging, multi-electrode extracellular recordings, patch-clamp recordings, holography based optogenetics and psychophysics in awake behaving animals, our goal is to characterize in detail single-cell and neural network activity in the rodent cortex during the learning process of a decision making task.

The Guermonprez lab investigates the cellular basis of anti-tumor immunity. We intend to understand cellular cross-talks between dendritic cells and anti-tumoral T lymphocytes in the context of cancer development and immunotherapies. The main objectives of this project are:
– to delineate myeloid cells and T cell infiltrate diversity in preclinical models of cancer and clinical samples defining immune landscapes,
– to understand how myeloid cells impact on T cell localization within tumors,

A significant proportion of patients with COVID-19 still experiences symptoms more than 3 months after an initial infection with SARS-CoV-2. Persisting or reemerging symptoms are varied, with a predominance of asthenia, neurocognitive impairment, and dyspnea. The persistence of these debilitating symptoms defines the Long COVID syndrome, which is now considered a serious public health concern. Importantly, the etiology of Long COVID remains unknown. We propose to test the hypothesis that an inefficient antiviral T cell response underlies Long COVID. The rationale is that suboptimal T cell immunity could promote viral persistence, with residual virus in turn causing localized tissue damage and persistent inflammation.

Lung tissue resident memory CD8+ T cells (TRMs) represent a population of cytotoxic lymphocytes able to provide frontline immunity against respiratory infections. There is strong evidence that lung cancer (NSCLC) infiltration by CD8+ TRMs is associated with favourable clinical outcomes and improved responses to immune checkpoint blockade immunotherapies. Despite their clinical relevance, the mechanisms underlying the activation and maintenance of TRMs remain ill-defined. The main objective of this project is to understand the impact of dendritic cell populations on the infiltration of lung cancer by CD8+ TRMs during lung cancer development.

We are looking for a highly motivated researcher willing to explore the role of 1D wetting in topography sensing, by developing 3D in vitro models in collaboration with the laboratories headed by Sylvie Coscoy at Institut Curie and Vincent Semetey at Chimie Paristech. Two-photon induced polymerization of photoactivable resin will be used to form adhesive fibers of different shapes and sizes. Cellular behavior on these 3D microstructures will be observed and quantified and the mechanisms leading to the remodeling of the actin cytoskeleton will be determined. We will also assess how this topography sensing process impacts essential cellular processes, such as cell motility and interaction of bacteria with host cells. Overall, this project aims at understanding how cells navigate in the complex environment formed by the extracellular matrix and how pathogenic bacteria take advantage of this process during infection.

Our laboratory has one position available for postdoctoral fellow in the area of inflammation and regenerative disease research. We are interested in mechanisms of tissue homeostasis, adaptation and repair, with an emphasis on tissue long lived cells such as stromal cells. Using novel genetic and pharmacological tools, we investigate the stromal signaling and crosstalk with immune cells, stem cells and other resident cells in different physiological and pathological conditions with a focus on inflammatory and regenerative disorders. We perform mostly in vivo studies, combining lineage tracing and depletion models, flow cytometry, confocal microscopy and unbiased approaches such as single cells transcriptomics and epigenetics to characterize molecular and cellular mechanisms.
