The long-term aim of research in the Lehrbach lab is to better understand proteostasis mechanisms and their roles in aging, neurodegenerative diseases and cancer. The fellow will work on individual and collaborative projects that use genetic, genomic, and cell and molecular biology approaches to discover new proteostasis mechanisms and reveal their physiological roles. The primary model organism for this work will be C. elegans, however previous experience with C. elegans is not essential.

A postdoctoral fellow position in Neuroscience/Neurodegeneration is available in the Fred Hutch lab of Dr. Linda Buck. Dr. Buck received a Nobel Prize in Physiology or Medicine in 2004 for “Odorant receptors and the organization of the olfactory system”. Current projects in the Buck lab focus on molecular mechanisms that underlie stress, appetite, and neurodegeneration and how they are influenced by the olfactory system. The lab has developed and uses cutting edge technologies to unravel neural mechanisms and neural circuits underlying specific functions/dysfunctions. These include novel genomics methods to define neural circuits, genetic and chemical control of neuronal activation, imaging of gene expression and neuronal activation, gene editing, and single cell genomics.

Our studies utilize human immunology, murine models of cancer, state-of-the-art imaging techniques, RNA sequencing techniques, and immunological techniques to explore the role of the innate immune system in controlling immune responses to cancer. A major focus of our lab is to study how B cells and the autoantibodies they produce recognize and target tumor antigens. We’ve demonstrated tumor-specific autoantibodies can be excellent biomarkers of lung tumorigenesis and can be detected prior to diagnosis (see our recent publications in AJCCRM 2019, JNM 2022 and STM 2023, PUBMED#’s 30422669, 35483965, 36630482). Using our unique approach of capturing autoantibodies bound to their native tumor derived epitopes ex vivo, we’ve shown tumor-specific autoantibodies target posttranslational modifications with high specificity. With this knowledge, we’ve isolated posttranslational tumor-specific B cells directly from patients. Our goal is to translate these findings into antibody-based theranostic applications.

The Ring Lab of Synthetic and Systems Immunopharmacology is located within the Translational Science and Therapeutics Division and is a member of the Immunotherapy Integrated Research Center (IIRC). Their primary focus is on developing unique and first-in-class therapeutic drugs for cancer immunotherapy using cutting-edge protein engineering with directed evolution. The lab has developed therapeutics that are currently in clinical trials, such as ST-067 (a “decoy-resistant” IL-18), MDNA-11 (CD122-biased IL-2), and ALX-148/evorpacept (an ultra-high affinity SΙRPα decoy). The Newell Lab is a highly collaborative group within the Vaccine and Infectious Disease Division of the Fred Hutchinson Cancer Center. The lab focuses on the application and development of novel approaches for studying antigen-specific T cells in the context of cancer and infectious diseases. In addition to leveraging expertise in the use of highly-multiplexed peptide-MHC tetramer staining in conjunction with mass cytometry to deeply profile antigen-specific T cells, novel single cell sequencing and computational approaches are being used that allow for improved examination of T cell phenotypes, function, antigen specificity, as well as T cell receptor repertoire. Although the lab focuses mostly on studying the roles of T cells in human health and disease, mouse tumor systems are also employed. A major goal of lab is to identify useful biomarkers of clinical outcomes that take advantage of modern experimental and computational approaches with particular focus on human infectious diseases and cancer.
