The Biosphere Theory and Modeling (BTM) group investigates how the Earth functions as a whole system, the role of life within it, and what a sustainable human future might look like. To do this, we take a unique Earth system approach that focuses on energy and thermodynamics. Previous work by our group has shown that thermodynamics and its fundamental limits strongly determine the dynamics of the Earth system, and thus the exchanges between land, ecosystems, and the atmosphere. This can be used to construct very simple yet physically-based models that reproduce observations very well, setting a basis to evaluate land-atmosphere exchange and interactions, the function of the hydrologic cycle, the role of vegetation, and climate change sensitivities.

Single molecule X-ray scattering experiments with free electron lasers offer a new route to the structure determination of biomolecules. In addition, they hold the potential to not only determine the molecular structure, but also its dynamics. However, computing the structure from such data poses a highly complex mathematical problem, and new theoretical methods and algorithms are required. The main goal of this particular research project is to develop Bayesian methods to determine molecular structures and dynamics. The influence of peers is one of the most important drivers of juvenile delinquency. Several decades of research have taught us that having delinquent friends increases the risk of becoming delinquent oneself and that spending time with delinquent peers increases the likelihood of committing crimes of one’s own. These findings hold true across culture, context, and research method. However, prior research has principally relied on introspective and retrospective research methods, such as interviews and panel studies. Exactly how peers influence each other, i.e., what happens in the moment of decision making, is less well understood. Probably no more than a handful of criminological studies have addressed this question experimentally.

The DMP group at MPI Magdeburg is an inter-disciplinary research unit addressing these questions from different perspectives. The theoretical centerpiece of our work is the Koopman operator description of dynamical systems, which leads to a linearization of a dynamical system using an infinite-dimensional feature space. On a theoretical level, we are interested in describing the convergence and approximation error of data-driven approximations to the Koopman operator. About Postdoctoral Position

We are interested in revealing the molecular secrets of ADP-ribosylation (ADPr), a challenging PTM that plays key roles in health and disease. Using advanced proteomics, we uncovered Serine ADPr as a new type of histone mark (Nature Chemical Biology 2016) and described the underlying molecular mechanisms by identifying HPF1/PAPR1 as its reader complex (Mol Cell 2017) and ARH3 as its eraser (eLife 2017). Crucially, we and others have shown that Ser-ADPr is the primary form of ADPr in the DNA damage response (eLife 2018).

These insights are stimulating studies of how Ser-ADPr regulates the DNA damage response and chromatin dynamics, as exemplified by our recent report on the interplay between Ser-ADPr and canonical histone marks (Cell Reports 2018). Recently we established a chemical biology strategy that is based on the Ser-ADPr writer complex and allows the generation of a range of essential tools for ADPr. This novel technology has broad potential to illuminate ADPr signalling pathways in essential biological processes and has allowed us to elucidate the prevalence of mono-ADPr upon DNA damage (Cell 2020).

A multitude of DNA-repair pathways ensure genomic integrity in a healthy cell. Disbalances or dysfunctionalities in these pathways can lead to genomic instability, which is a central contributor to carcinogenesis and ageing. Our research aims to provide a better understanding of the relationship between genome maintenance, ageing, and cancer, in an interdisciplinary approach. Within this project, we dissect defects in genome instability in patients suffering from mantle cell lymphoma, a mature B-cell lymphoma typically affecting the elderly. For these patients, mechanism-based therapies are scarce and treatment outcomes remain relatively poor. We investigate the mechanisms underlying this disease and its tumor microenvironment composition to identify potential novel therapeutic approaches.
